The nuclear protooncogenes c-jun and c-fos as regulators of DNA replication.

چکیده
Polyomavirus (Py) DNA replication is regulated by its enhancer, which contains an AP1 site, c-Jun and c-Fos, the products of nuclear protooncogenes c-jun and c-fos, form the heterodimeric transcriptional activating factor AP1. Overexpression of c-fos and c-jun genes strongly stimulated Py DNA replication from the Py origin of replication as well as transcription from the Py early promoter through the AP1 binding site. The cAMP response element (CRE)-binding protein CREB stimulated only transcription, not DNA replication, through the CRE under similar conditions. The results indicate that AP1 functions as a regulator of DNA replication and that the mechanism of activation of Py DNA replication by AP1 is distinct from that of activation of transcription from the Py early promoter.

A ubiquitous nuclear protein stimulates the DNA-binding activity of fos and jun indirectly.
The protooncogenes c-fos and c-jun encode nuclear proteins (fos and jun, respectively) that function cooperatively as a heterodimeric protein complex in the regulation of gene transcription. These proteins dimerize via a structural motif known as the leucine zipper and bind to activator protein-1 sites via a conserved domain that is rich in basic amino acids. Previously, we demonstrated that wh...
متن کاملThe effects of gestational and lactational exposure to Nonylphenol on c-jun, and c-fos expression and learning and memory in hippocampus of male F1 rat
Objective(s): To investigate the effects of Nonylphenol (NP) in pups from dams exposed during gestational and lactational periods on immediate early genes (c-jun, c-fos) in hippocampus and the learning and memory of F1 rats. Materials and Methods: Twenty eight pregnant dams, stratified by pregnancy date, were randomly assigned into 4 groups, which were gavaged with NP at the doses of 50 mg/kg/d...
متن کاملEffect of trichloroethylene on DNA methylation and expression of early-intermediate protooncogenes in the liver of B6C3F1 mice.
Trichloroethylene (TCE) is a multimedia environmental pollution that is carcinogenic in mouse liver. The ability of TCE to modulate DNA methylation and the expression of immediate-early protooncogenes was evaluated. Female B6C3F1 mice were administered 1000 mg/kg TCE by gavage 5 days/week and killed after 5, 12, or 33 days of exposure. Methylation of DNA as 5-methylcytosine was decreased by 5 d...
متن کاملDifferential induction of c- fos, c- jun, and apoptosis in lung epithelial cells exposed to ROS or RNS.
Reactive oxygen (ROS) or nitrogen (RNS) species can affect epithelial cells to cause acute damage and an array of pulmonary diseases. The goal of this study was to determine patterns of early response gene expression and functional end points of exposure to nitric oxide (NO ⋅), H2O2, or peroxynitrite (ONOO-) in a line of rat lung epithelial (RLE) cells. Our focus was on c- fos and c- jun protoo...
متن کاملEstradiol-17beta stimulates proliferation of mouse embryonic stem cells: involvement of MAPKs and CDKs as well as protooncogenes.
Although the importance of estradiol-17beta (E(2)) in many physiological processes has been reported, to date no researchers have investigated the effects of E(2) on embryonic stem (ES) cell proliferation. Therefore, in the present study, we have examined the effect of E(2) on the DNA synthesis of murine ES (ES-E14TG2a) cells and its related signaling pathways. The results of this study show th...
